چکیده

(+)-Amphetamine sulphate infused directly into the substantia nigra, pars compacta, at low rates and in low volumes and concentrations, produces a marked and prolonged inhibition of neuronal activity. This effect was significantly reduced or abolished following (+/-)-alphamethyl-p-tyrosine pretreatment or following electrolytic lesions in the diencephalon anterior to the substantia nigra. These results confirm previous reports of similar effects of such treatments on the depression of neuronal firing produced by systemically administered amphetamine. Further, our results suggest that (+/-)-alphamethyl-p-tyrosine and anterior diencephalic lesions are affecting an indirect dopaminergic inhibitory mechanism intrinsic to the substantia nigra. Such evidence may also be regarded as consistent with the view that amphetamine produces an inhibition of dopaminergic neuronal activity by affecting release of dopamine from dendrites of dopaminergic neurones, i.e. by affecting self-inhibition by dopaminergic neurones.

Involvement of dopaminergic mechanisms in racemate pentazocine induced stereotyped behaviour in rats.

Racemate pentazocine was found to induce stereotyped behaviour (SB) in rats. Pretreatment with haloperidol and alpha-methyl-p-tyrosine significantly antagonised dl-pentazocine induced SB. This indicates that dl-pentazocine induces SB by releasing dopamine (DA) from the nigrostriatal and mesolimbic dopaminergic neurones with resultant activation of the postsynaptic striatal and mesolimbic D2 DA ...

Dopamine depletion does not protect against acute 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine toxicity in vivo.

Dopamine (DA) has been postulated to play a role in the loss of dopaminergic substantia nigra (SN) neurons in Parkinson's disease because of its propensity to oxidize and form quinones and other reactive oxygen species that can alter cellular function. Moreover, DA depletion can attenuate dopaminergic cell loss in vitro. To test the contribution of DA to SN impairment in vivo, we used DA-defici...

Prevention of 1-methyl-4-phenylpyridinium- and 6-hydroxydopamine-induced nitration of tyrosine hydroxylase and neurotoxicity by EUK-134, a superoxide dismutase and catalase mimetic, in cultured dopaminergic neurons.

Oxidative stress has been implicated in the selective degeneration of dopaminergic (DAergic) neurons in Parkinson's disease (PD). In this study, we tested the efficacy of EUK-134, a superoxide dismutase (SOD) and catalase mimetic, on the nitration of tyrosine hydroxylase (TH), a marker of oxidative stress, and neurotoxicity produced by 1-methyl-4-phenylpyridinium (MPP(+)) and 6-hydroxydopamine ...

MORPHINE AND APOMORPHINE INHIBIT GASTROINTESTINAL TRANSIT (GIT) THROUGH TWO DIFFERENT MECHANISMS

Morphine was used as a remedy for the control of diarrhea centuries before it's sedative-analgesic effect was discovered. Although several mechanisms have been proposed for the morphine-induced inhibition of gastrointestinal transit (OIT), the exact mechanism has not yet been identified. On this basis the possible involvement of the dopaminergic system in morphine-induced inhibition of tra...

Oxidative stress and mitochondrial-mediated apoptosis in dopaminergic cells exposed to methylcyclopentadienyl manganese tricarbonyl.

Methylcyclopentadienyl manganese tricarbonyl (MMT), an organic manganese-containing gasoline additive, was investigated to determine whether MMT potentially causes dopaminergic neurotoxic effects. MMT is acutely cytotoxic and dopamine-producing cells (PC-12) seemed to be more susceptible to cytotoxic effects than nondopaminergic cells (striatal gamma-aminobutyric acidergic and cerebellar granul...
